Target Background

Function
Poly-ADP-ribosyltransferase that mediates poly-ADP-ribosylation of proteins and plays a key role in DNA repair. Mediates glutamate, aspartate or serine ADP-ribosylation of proteins: the ADP-D-ribosyl group of NAD(+) is transferred to the acceptor carboxyl group of target residues and further ADP-ribosyl groups are transferred to the 2'-position of the terminal adenosine moiety, building up a polymer with an average chain length of 20-30 units. Mainly mediates glutamate and aspartate ADP-ribosylation of target proteins in absence of CG1218/HPF1. Following interaction with CG1218/HPF1, catalyzes serine ADP-ribosylation of target proteins; CG1218/HPF1 conferring serine specificity by completing the Parp active site.; Plays a fundamental role in organizing chromatin on a global scale. Autoregulates Parp transcription by influencing the chromatin structure of its heterochromatic environment.
Gene References into Functions
  1. The results demonstrate that PARylation process in Drosophila is tightly regulated in the context of strands-breaks repair; PARP is essential during the maintenance of DNA integrity, but dispensable in the DNA repair process. PMID: 27793508
  2. A mutation of Parp also increases NAD+ levels; although, this was only observed in parkin mutant flies and not in the heterozygous Parp mutants, possibly owing to an increased PARP activity in the parkin mutants. PMID: 27031963
  3. Based on these findings, we propose a model that explains how PARP1 activity impacts nucleolar functions and, consequently, ribosomal biogenesis PMID: 22242017
  4. PARP is associated with the 5' end of Hsp70, and its enzymatic activity is rapidly induced by heat shock leading to nucleosome loss. PMID: 22178397
  5. Activation of PARP-1 overexpression in the imago results in extension of the lifespan in females and males. The lifespan increase in females with PARP-1 conditional overexpression was accompanied by decrease of fertility. PMID: 22184968
  6. inhibits transcription by contributing to the condensation of chromatin, which creates a barrier against gene transcription PMID: 20591646
  7. demonstrate that this alteration specifically excludes PARP1 protein from heterochromatin and makes PARP1 unable to maintain repression of retrotransposable elements. PMID: 20371698
  8. PARP-e autoregulates Parp transcription PMID: 12183365
  9. propose that chromosomal PARP molecules become activated by developmental or environmental cues and strip nearby chromatin proteins off DNA to generate a puff PMID: 12543974
  10. PARP1 is required for protein targeting to Cajal body. PMID: 19229318
