Description

This human IL1A ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human IL1A in multiple samples, including serum, plasma, cell culture supernates, tissue homogenates, or cell lysates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of IL1A bound in the initial step.

IL1A is constitutively expressed in epithelial, endothelial, and stromal cells and can be upregulated in hematopoietic and non-hematopoietic cells by diverse stimuli, including TLR agonists, inflammatory cytokines, and oxidative stress. In non-hematopoietic cells, IL1A signals via IL1R on myeloid cells in a MyD88/RIPK1/(TAK1)-dependent manner to induce the expression of pro-inflammatory cytokines. IL1A also plays a role in neutrophil recruitment and host defense after infections caused by viral, fungal, or protozoan. Dysregulation of IL1A production is associated with numerous auto-inflammatory disorders.

 Q&A
Q:

I am interested in ELISA Kit Cat#CSB-E04620h. My highest concentration of testing drug will contain 10% DMSO. I am worried the high concentration DMSO will affect the ELISA. Could you help to provide information about my worries? Thank you for your help.
My Experiment Procedure:
1. culture cell
2. Treat testing drug
3. Use supernates to run ELISA

A:
Thanks for your inquiry. DMSO indeed will affect elisa assay as it will react with hydrophobic group of protein and change the protein. We suggest you do a pretest first.

Target Background

Function
(From Uniprot)
Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.
