To assess the linearity of the assay, samples were spiked with high concentrations of mouse MMP-9 in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.
These standard curves are provided for demonstration only. A standard curve should be generated for each set of samples assayed.
The mouse MMP9 ELISA Kit is engineered for accurate measurement of mouse MMP9 levels from samples including serum, plasma, or tissue homogenates. It uses the Sandwich-ELISA mechanism in combination with the enzyme-substrate chromogenic reaction to measure the MMP9 content in the sample. The color intensity is positively correlated with MMP9 content in the sample. This kit has been validated against standards of sensitivity, specificity, precision, linearity, recovery, and lot-to-lot consistency.
MMP9 is a secreted zinc-dependent extracellular endopeptidase responsible for the cleavage of extracellular matrix (ECM) proteins, creating permissive environments for neuronal development and plasticity. It is primarily secreted by neutrophils and macrophages and is involved in the regulation of inflammation in tissue and diseases. MMP9 is also associated with periodontal inflammation and is remarkably increased in chronically infected areas in apical periodontitis. It also mediates and facilitates bone destruction. MMP9 expression declined osteoclast formation and suppressed pro-inflammatory cytokines secretion. In pathophysiological conditions, MMP9 is upregulated during development and wound healing, as well as during pathologies involving inflammatory processes, including diabetes, arthritis, and cancer.

Review: We used CSB-E08007m to detect MMP-9 in normal mouse skin tissue homogenate, and found that its ODA450 value was 2.28 when the homogenate stock solution was used, 1.227 when the homogenate was diluted twice, 0.818 when it was diluted four times, and 0.6215 when it was diluted eight times, and then it was calculated that its concentrations corresponded to 48.98, 25.50, 16.68, and 12.53 ng/mL respectively when using the stock solution, which exceeded the detection range of the kit by 0.625-40 ng/mL. After calculating its concentration corresponded to 48.98,, 25.50, 16.68, 12.53 ng/mL, respectively, and found that the use of the original solution exceeded the detection range of the kit by 0.625-40 ng/mL, so we finally determined to dilute the homogenate by 5-fold, and then carry out the following groups of tests. The kit has a wide detection range, high sensitivity, high precision, easy operation, good fit of the calibration curve, very easy to start, and the results are accurate and reliable.
