CUSABIO's IL1B rat ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of rat IL1B in serum, plasma, or tissue homogenates. It has good reproducibility, high specificity & sensitivity, and good linearity. This assay uses an antibody specific for rat IL1B coated on a 96-well plate. IL1B present in a sample is bound to the wells by the immobilized antibody after adding the sample into the wells. Biotinylated IL1B antibody is added to the wells, forming an antibody-analyte-antibody complex. HRP-conjugated avidin is pipetted to the wells and binds to the complex. A TMB substrate solution is added to the wells and color develops in proportion to the amount of IL1B bound. The addition of Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm using a microplate reader. IL1B is a potent pro-inflammatory cytokine essential for host-defense responses to infection and injury. IL1B binds to IL1RI and then associates with the IL-1RAcP, initiating the recruitment of the MyD88 adaptor and activation and ligation to IRAK4, activating the MAPK p38 cascade and the NF-κB signaling pathways that are implicated in inflammatory responses. It is involved in pain, inflammation, and autoimmune disorders. IL1B also exerts important homeostatic effects in the normal organism, including the modulation of temperature, feeding, and sleep. Aberrant expression of IL1B has been found in various fibrotic diseases. For example, IL1B overexpression participates in the pathophysiological changes that occur during different disease states, such as rheumatoid arthritis, osteoarthritis, and Alzheimer's disease.
