IL-18 plays a central role in Th1 polarization by synergizing with IL-12 to drive IFN-γ production, yet its activity is notoriously sensitive to endotoxin contamination that can confound immune cell readouts. This tag-free recombinant human IL-18, spanning the full mature protein sequence (aa 37–193), demonstrates potent bioactivity with an ED50 ≤2.0 ng/mL in inducing IFN-γ secretion by KG-1 cells co-stimulated with TNF-α, providing a quantitative benchmark for dose-response studies in Th1 differentiation and macrophage activation assays. Endotoxin levels ≤10 EU/mg satisfy the stringent criteria expected for primary immune cell cultures, PBMC-based functional assays, and in vivo inflammation models where LPS artifacts must be minimized. Purity exceeding 95% by SDS-PAGE, combined with the validated signaling potency, supports use in JAK/STAT and NF-κB pathway analysis, as well as antibody development workflows requiring a functional immunogen or ELISA standard.
