The recombinant human IL18 protein is encoded by a recombinant DNA that was cloned into the expression vector and then transfected into the yeast cells that support the expression of the gene. The recombinant DNA was constructed by fusing the N-terminal 10xHis tag gene to the gene fragment coding for the 1-193aa of the human IL18 protein. After purification, the product is the recombinant human IL18 protein. This recombinant IL18 protein was subjected to the SDS-PAGE determination. Its purity reaches over 90% evaluated by Bandscan software analysis combined with SAS-PAGE. This IL18 protein ran to the molecular weight of about 25 kDa under SDS-PAGE condition. It may find use in immunology research.
Interleukin-18 (IL-18) belongs to the IL-1 family of cytokines. The inactive IL-18 precursor is synthesized as its active form by caspase-1. In humans, increased disease severity can be associated with an imbalance of IL-18. IL-18 has been found to be associated with the injury of different organs. Furthermore, the overly imbalance of cytokines could cause potentially fatal conditions such as a cytokine storm. IL-18 forms a signaling complex by binding to the IL-18 alpha chain. It has been reported that IL-18 is associated with hyperphagia and a variety of metabolic syndrome. Importantly, increasing numbers of animal and clinical studies indicate a role for IL-18 in heart disease.
