TGF-β1 orchestrates a broad spectrum of cellular responses—from epithelial-mesenchymal transition to immune suppression—making precise control of its bioactivity essential in mechanistic studies of fibrosis, oncogenesis, and tissue repair. This mammalian-expressed fragment spanning residues 279–390 represents the mature, bioactive domain and demonstrates potent growth-inhibitory activity with an ED50 ≤0.1 ng/mL in TF-1 cell proliferation assays conducted in the presence of IL-4, confirming receptor engagement at physiologically relevant concentrations. Mammalian expression ensures native disulfide bonding and folding architecture critical for high-affinity receptor binding, supporting its use in Smad phosphorylation assays, epithelial-mesenchymal transition models, and receptor competition studies by surface plasmon resonance or ELISA. Purity exceeding 95% by SDS-PAGE and endotoxin levels ≤10 EU/mg satisfy the quality thresholds commonly required for primary cell differentiation experiments, including osteogenic and chondrogenic lineage commitment assays, as well as for in vivo wound healing and tumor microenvironment models where low endotoxin is essential to avoid confounding inflammatory signals.

Associates non-covalently with TGF-beta-1 and regulates its activation via interaction with 'milieu molecules', such as LTBP1, LRRC32/GARP and LRRC33/NRROS, that control activation of TGF-beta-1. Interaction with LRRC33/NRROS regulates activation of TGF-beta-1 in macrophages and microglia (Probable). Interaction with LRRC32/GARP controls activation of TGF-beta-1 on the surface of activated regulatory T-cells (Tregs).
