TGF-β1 orchestrates a broad spectrum of cellular responses—from epithelial-to-mesenchymal transition to immune suppression—making precise control of its bioactivity essential in mechanistic studies of fibrosis, cancer progression, and stem cell fate decisions. This preparation spans residues 279–390, encompassing the mature bioactive domain, and demonstrates potent growth-inhibitory activity with an ED50 ≤0.1 ng/mL in TF-1 cell proliferation assays conducted in the presence of IL-4, confirming receptor engagement at physiologically relevant concentrations. Mammalian expression preserves the native disulfide architecture and any post-translational modifications critical for high-affinity receptor binding, supporting its use in stem cell differentiation protocols, receptor-ligand competition assays by surface plasmon resonance or ELISA, and in vitro wound healing models where TGF-β1 signaling drives myofibroblast activation. Purity exceeding 95% by SDS-PAGE and endotoxin levels ≤10 EU/mg satisfy the quality thresholds commonly required for cell-based functional assays and antibody characterization workflows.

Associates non-covalently with TGF-beta-1 and regulates its activation via interaction with 'milieu molecules', such as LTBP1, LRRC32/GARP and LRRC33/NRROS, that control activation of TGF-beta-1. Interaction with LRRC33/NRROS regulates activation of TGF-beta-1 in macrophages and microglia (Probable). Interaction with LRRC32/GARP controls activation of TGF-beta-1 on the surface of activated regulatory T-cells (Tregs).
