The major mite allergen Der p 23 polyclonal antibody production process involves the selection of a recombinant Dermatophagoides pteronyssinus major mite allergen Der p 23 protein segment (amino acids 22-90) as the immunogen. This protein is used to immunize a rabbit, generating an antibody response. Following immunization, rabbit serum is collected to obtain polyclonal antibodies. These antibodies are then subjected to affinity-chromatography purification, where they are specifically isolated based on their reactivity to the major mite allergen Der p 23 protein. Subsequently, the functionality of the antibody is assessed in ELISA and WB assays, demonstrating its suitability for detecting Dermatophagoides pteronyssinus major mite allergen Der p 23 protein in experimental applications.
The group 23 allergen, Der p 23, from Dermatophagoides pteronyssinus, is a peritrophin-like protein that contains a chitin-binding domain in the sequence but does not bind chitinis. It is a major allergen amongst house dust mite (HDM)-sensitized individuals and is present in the outer membrane of mite feces. Der p 23 has high IgE binding frequency and high allergenic activity. It can induce persistent respiratory symptoms.
