CRLF2 forms a heterodimeric receptor complex with IL-7Rα to mediate thymic stromal lymphopoietin (TSLP) signaling, a pathway implicated in allergic inflammation and certain B-cell acute lymphoblastic leukemias. This biotinylated extracellular domain (aa 25–231) is expressed in mammalian cells to preserve native glycosylation and disulfide architecture, yielding a protein that demonstrates quantifiable antibody binding in functional ELISA with an EC50 of 3.631–7.668 ng/mL when immobilized on streptavidin-coated surfaces. The streptavidin-compatible biotin tag enables oriented capture in surface plasmon resonance and biolayer interferometry experiments for affinity characterization of therapeutic antibody candidates targeting CRLF2, as well as competitive inhibition assays to screen blocking antibodies or TSLP antagonists. Purity exceeding 95% by SDS-PAGE and endotoxin below 1.0 EU/μg align with standards expected in antibody epitope mapping and ligand-receptor interaction studies where low background and minimal inflammatory artifacts are essential.
