CRLF2 forms a heterodimeric receptor complex with IL-7Rα to mediate thymic stromal lymphopoietin (TSLP) signaling, a pathway central to allergic inflammation and lymphoid development. This mammalian-expressed construct spanning residues 25–231 captures the complete extracellular ligand-binding domain and demonstrates quantified binding to human TSLP with an EC50 of 41.01–45.10 ng/mL in functional ELISA, providing a validated reagent for ligand-receptor interaction studies and competitive inhibition assays. The native post-translational modifications conferred by mammalian expression support conformational integrity critical for epitope mapping of therapeutic antibodies targeting the TSLP–CRLF2 axis, as well as for screening biologics or small-molecule inhibitors in drug discovery campaigns. With purity exceeding 95% by SEC-HPLC and endotoxin levels below 1.0 EU/μg, this protein meets the quality thresholds commonly required for surface plasmon resonance affinity characterization, biolayer interferometry kinetics studies, and use as a positive control in receptor-binding platforms.
