CRLF2

CDLF2, also referred to as CRLF2, belongs to the type I cytokine receptor family. This protein acts as a receptor for thymic stromal lymphopoietin (TSLP) and, when paired with the interleukin-7 receptor (IL7R), activates the STAT3, STAT5, and JAK2 pathways. These pathways are key to cell growth and the development of the hematopoietic system. CRLF2's action is through forming complexes with TSLP and IL7R, which then activate the JAK-STAT signaling pathway to stimulate cell proliferation. CRLF2 is essential for the normal development of B lymphocytes. However, in acute lymphoblastic leukemia (ALL), especially the Ph-like ALL. CRLF2 overexpression or rearrangement is associated with disease development. Due to genetic rearrangements, upregulated CRLF2 expression on pre-B lymphocytes can drive excessive cell proliferation, which is also a marker for a less favorable prognosis.
