Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description
Recombinant Human Hyaluronidase PH-20(SPAM1) is a partial length protein expressed with N-terminal 6xHis-SUMO-tagged in the E.coli. Its expression region corresponds to 36-490aa of human SPAM1 protein. Its purity was determined by SDS-PAGE and reached up to 90% and presented a molecular mass band of 67.9 kDa on the gel. This recombinant SPAM1 protein may be used to synthesize antibodies against SPAM1.
SPAM1 is involved in sperm-egg adhesion. Upon fertilization, sperm must first penetrate a layer of cumulus cells that surrounds the egg before reaching the zona pellucida. The cumulus cells are embedded in a matrix containing hyaluronic acid which is formed prior to ovulation. This protein aids in penetrating the layer of cumulus cells by digesting hyaluronic acid. Diseases associated with SPAM1 include Mucopolysaccharidosis, Type Ix and Aqueous Misdirection.

 Q&A
Q:

Please send me a quotation for the following inquiry.
I want to Mammalian cell expression for CHO-S cell.
I am trying to make a cloning in the HZ24 plasmid containing 5 NheI and 3 BamHI restriction sites.
Please refer to below link that is desired protein sequence.

The patents in the link below shows how cloning and expression are performing in full details.

It cannot be necessarily the same, just refer to reference, and trying to make active PH20 soluble for expression after glycosylation.

A:
Very nice to receive your inquiry. Your specified sequence is only a few amino acids shorter than the mature full length protein, so we recommend to express the full length of the mature protein.

You can view the full information of sequence and tag on this page.
Sorry, we didn't check the corresponding product on R&D website, in theory, the protein expressed by the Mammalian cell system should have the post-translational modification, however, we haven't done any validation.
Please remark your requirement for tag removal if you need the untagged protein or tag removal when placing the order.
Sorry, we haven't tried to remove the tag before. We can try enzyme digestion, but we can't guarantee 100% successfully.
The overall success rate of enzyme digestion data analysis is 75%-86%.
Not all protein tags can be removed as some proteins will be very unstable after tag removal.
If we succeed in removing the tag, we will charge for extra cost.
If we fail in removing the tag, we won’t charge for tag removal and provide the fusion protein, and remark this information in datasheet as follows
“Note: The laboratory determined that the Tag on your protein could not be removed with standard laboratory procedures. Your protein is being supplied with the Tag intact.”
Generally, the delivery time will be extended for 3 days.

Function
Involved in sperm-egg adhesion. Upon fertilization sperm must first penetrate a layer of cumulus cells that surrounds the egg before reaching the zona pellucida. The cumulus cells are embedded in a matrix containing hyaluronic acid which is formed prior to ovulation. This protein aids in penetrating the layer of cumulus cells by digesting hyaluronic acid.
