To generate the biotinylated recombinant human tyrosine-protein phosphatase non-receptor type substrate 1 (SIRPA) in mammalian cells, first, construct an expression vector containing the target gene fused with a 6xHis-Avi-tag sequence at the C-terminus. The target gene encodes the amino acids 31-370 of the human SIRPA protein. And then, transfect the recombinant vector into mammalian cells using appropriate methods. Culture the transfected cells and induce expression of the recombinant SIRPA protein. Introduce a biotin ligase enzyme or co-express it with the recombinant protein to attach biotin molecules to the 6xHis-Avi-tag sequence. Harvest the cells or collect the culture supernatant and purify the biotinylated recombinant SIRPA protein. The purity of the biotinylated recombinant human SIRPA protein is over 90%.
