Using a synthesized peptide derived from human JAK1 as an immunogen, B cells were obtained and fused with myeloma cells to form hybridomas. The hybridomas' variable light (VL) and variable heavy (VH) domains' cDNA were sequenced, and the sequencing data was used to generate a recombinant vector containing the JAK1 monoclonal antibody gene. This recombinant vector was transfected into cells for culture, and the JAK1 recombinant monoclonal antibody was collected and purified from the cell culture supernatant using affinity chromatography. The purified antibody was then tested for specificity in ELISA and IHC applications. This antibody exclusively detects human JAK1 protein.
The JAK1 protein is a member of the JAK family of non-receptor tyrosine kinases. It plays a crucial role in signal transduction pathways that are activated by various cytokines and growth factors. JAK1 is involved in a wide range of cellular processes including cell growth, proliferation, differentiation, survival, and immune response. In addition to its role in cytokine signaling, JAK1 is also involved in the regulation of cellular adhesion and migration.
