JAK1, or tyrosine kinase 2 subfamily member 1, alias Janus kinase 1. JAK1 is a non-receptor tyrosine kinase belonging to the Janus kinase family that is widely involved in intracellular signaling. It is encoded by its own gene in human cells and is expressed in a wide range of cell types.The mechanism of action of JAK1 is mainly through interaction with the intracellular portion of cell-surface receptors.When these receptors are activated by the corresponding ligands, JAK1 is recruited and activated, which in turn initiates the downstream signaling pathways by autophosphorylating and phosphorylating tyrosine residues of the receptors to provide binding sites for signaling molecules.
JAK1 is biologically significant and is involved in a variety of cell signaling processes, including cell proliferation, differentiation, apoptosis, and immune responses, etc. JAK1 is associated with a variety of cytokine receptors, such as interferon, growth factor, and hormone receptors, etc. By activating signaling at these receptors, JAK1 participates in the regulation of STAT (Signal Transducer and Activator of Transcription) protein By activating the signaling of these receptors, JAK1 is involved in the regulation of STAT (signal transducer and activator of transcription) protein phosphorylation and nuclear translocation, thereby affecting gene expression.Abnormal JAK1 function has been associated with a wide range of diseases, especially autoimmune diseases and cancers, and therefore the development of JAK1 inhibitors has become a new strategy for the treatment of these diseases.

IHC image of CSB-RA227790A0HU diluted at 1:100 and staining in paraffin-embedded human colon cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
