CYP27A1, a cytochrome P450 family enzyme, performs key mitochondrial roles, notably the 27-hydroxylation of cholesterol in liver bile acid production, providing an alternative pathway for bile acid synthesis. It also produces 27-hydroxycholesterol, which activates liver receptors and promotes cholesterol efflux from macrophages via ABCA1 and ABCG1, crucial for cholesterol transport and equilibrium. CYP27A1 is instrumental in vitamin D3 activation and is implicated in the development of several cancers, including those affecting the breast, prostate, colon, and liver.
