PDGFRA, platelet-derived growth factor receptor alpha, alias PDGFR-α, CD140A, etc.. It is a type I transmembrane receptor tyrosine kinase encoded by its own gene in human cells.The mechanism of action of PDGFRA is mainly through binding to its ligands platelet-derived growth factor AA (PDGF-AA), PDGF-AB, PDGF-CC, etc., which triggers receptor dimerization and autophosphorylation, and consequently activates downstream signaling pathways.Activation of PDGFRA promotes the proliferation, migration and survival of multiple cell types, including fibroblasts, pericytes and vascular smooth muscle cells, etc. PDGFRA has a wide range of biological significance, and it plays a key role in embryonic development, tissue repair, tumorigenesis and development, etc. Abnormal activation of PDGFRA signaling is closely related to the development of a variety of tumors, such as gastrointestinal mesenchymal stromal tumors and gliomas. In addition, PDGFRA is also involved in regulating angiogenesis and immune cell infiltration in the tumor microenvironment, affecting immune escape and drug tolerance in tumors. Therefore, PDGFRA is an important target in tumor therapy, and drugs such as small molecule inhibitors and monoclonal antibodies against PDGFRA have been used in clinical treatment.
