Because IL-17A levels can vary dramatically between healthy baseline and active disease states — and often fall in the low picogram range in circulation — accurate quantification demands both broad dynamic range and low-end sensitivity.
Detection Range: The 15.6–1000 pg/ml range spans from near-baseline serum IL-17A concentrations in healthy individuals to the elevated levels observed in active psoriasis, rheumatoid arthritis, and IBD flares. Most clinical and in vitro samples fall within this window without requiring extensive dilution, though stimulated Th17 culture supernates may need pre-dilution.
Sensitivity: At 3.9 pg/ml, the kit detects IL-17A in low-abundance contexts such as unstimulated PBMC conditioned media, early-stage autoimmune serum samples, or plasma from patients prior to disease flare, where circulating levels often hover in the single-digit picogram range.
Sample Compatibility: Validation across serum, plasma, cell culture supernates, and tissue homogenates enables researchers to correlate systemic IL-17A levels with local tissue expression and in vitro Th17 polarization output within a single experimental framework.
Assay Efficiency: The 1–5 hour protocol using 50–100 µl sample volumes accommodates limited clinical specimens and primary Th17 differentiation cultures where cell yields restrict supernate availability.
