The extracellular ligand-binding domain of human GLP1R (residues 24–145) is the critical region for incretin hormone recognition and a primary target for therapeutic antibody development in cardiometabolic research. This partial construct, expressed in mammalian cells to preserve native glycosylation and disulfide-mediated folding, demonstrates confirmed binding activity with an EC50 of 54.54–94.23 ng/mL against an anti-GLP1R recombinant antibody in functional ELISA, supporting its use in ligand-receptor interaction assays, competitive inhibition studies, and therapeutic antibody epitope mapping. The dual-tag design (N-terminal 10×His, C-terminal Myc) enables flexible oriented immobilization for SPR- or BLI-based affinity characterization and blocking antibody screening. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based binding assays and drug discovery workflows targeting GLP-1R-mediated signaling.
