  • Immunofluorescence staining of Hela cells with CSB-PA740922HA01HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).

Description

CUSABIO's product CSB-PA740922HA01HU is a polyclonal antibody against the CLEC9A protein. It was raised in the rabbit immunized with the recombinant human CLEC9A protein (57-241aa) and purified from rabbit serum by antigen affinity chromatography using the immunizing peptide. This CLEC9A antibody exists as the unconjugated IgG isoform. It was validated for use in ELISA, WB, and IF applications. It is reactive to human- and mouse-derived CLEC9A protein.

CLEC9A protein, also known as DNGR1, plays a critical role in recognizing dead cells and facilitating the processing and cross-presentation of dead cell-associated antigens in a Syk-dependent manner. The CLEC9A protein is also a key defining marker of the conventional DC1 (cDC1) subset. CLEC9A is a group V C-type lectin-like receptor that also functions as an activation receptor and can trigger proinflammatory cytokine generation.

Function
Functions as an endocytic receptor on a small subset of myeloid cells specialized for the uptake and processing of material from dead cells. Recognizes filamentous form of actin in association with particular actin-binding domains of cytoskeletal proteins, including spectrin, exposed when cell membranes are damaged, and mediate the cross-presentation of dead-cell associated antigens in a Syk-dependent manner.
