Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Description

Angiopoietin-2 functions as a context-dependent modulator of Tie2 signaling, making it a critical tool for investigating vascular remodeling, tumor angiogenesis, and endothelial cell survival. This recombinant human ANGPT2 covers the full mature sequence (aa 19–496) and is produced in mammalian cells, preserving native glycosylation patterns essential for proper folding and receptor-ligand interactions. Functional ELISA validation demonstrates strong binding potency, with an EC50 of 0.67–0.89 ng/mL against an anti-ANGPT2 recombinant antibody, supporting use as a reliable standard in antibody characterization assays, receptor-ligand binding studies by ELISA or SPR, and angiogenesis or wound healing models. Purity exceeding 90% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based proliferation and survival assays as well as in vivo tumor biology experiments where endotoxin contamination could confound results.

Function
Binds to TEK/TIE2, competing for the ANGPT1 binding site, and modulating ANGPT1 signaling. Can induce tyrosine phosphorylation of TEK/TIE2 in the absence of ANGPT1. In the absence of angiogenic inducers, such as VEGF, ANGPT2-mediated loosening of cell-matrix contacts may induce endothelial cell apoptosis with consequent vascular regression. In concert with VEGF, it may facilitate endothelial cell migration and proliferation, thus serving as a permissive angiogenic signal.
