Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

ENPP3 (CD203c) plays a critical role in purinergic signaling and has emerged as a target of interest in cancer biology, where its ectonucleotidase activity modulates the tumor microenvironment. This recombinant human ENPP3 construct, spanning residues 48–875 and expressed in mammalian cells, carries a C-terminal 10×His tag positioned to minimize interference with the catalytic domain, supporting its use in enzymatic activity assays, kinetic parameter analysis (Km, Vmax, kcat), and substrate specificity profiling. Functional validation by ELISA demonstrated specific antibody binding with an EC50 of 2.151–2.492 ng/mL, confirming proper folding and providing a suitable basis for inhibitor screening, IC50 determination, and deployment as a positive control in enzyme-linked assays. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typically required for accurate kinetic measurements and drug candidate evaluation studies.

Function

Hydrolase that metabolizes extracellular nucleotides, including ATP, GTP, UTP and CTP. Limits mast cell and basophil responses during inflammation and during the chronic phases of allergic responses by eliminating the extracellular ATP that functions as signaling molecule and activates basophils and mast cells and induces the release of inflammatory cytokines. Metabolizes extracellular ATP in the lumen of the small intestine, and thereby prevents ATP-induced apoptosis of intestinal plasmacytoid dendritic cells. Has also alkaline phosphodiesterase activity.
