ENPP3

Ectonucleotide pyrophosphatase/phosphodiesterase family member 3 is an enzyme that in humans is encoded by the ENPP3 gene. Hiroaki studied N2a cells in a 2013 experiment and purified an endogenous inhibitor present in GnT-IX, which was found to be identical to the external nucleotide pyrophosphatase/phosphodiesterase 3 (ENPP3), as demonstrated by mass spectral analysis and knock-out and overexpression of ENPP3 in cultured cells. Kinetic analysis showed that the ENPP3-induced inhibition of GnT-IX was mediated by ENPP3 in the hydrolysis of the nucleotide sugar donor substrate UDP-GlcNAc to produce UMP, a potent and competitive GnT-IX inhibitor. In fact, when ENPP3 is knocked out intracellularly, intracellular nucleotide sugar levels rise significantly and show changes in the characteristics of total cellular glycosylation. In addition to the partners of glycosyltransferases or other known modulators, ENPP3-mediated nucleotide hydrolysis in sugar will also have a broad and significant effect on glycosyltransferase activity and is responsible for altering total cellular glycosylation characteristics and regulating cell function.
