PVR (CD155/NECL-5) serves as a key ligand in the TIGIT immune checkpoint axis, making it a critical tool for dissecting co-inhibitory signaling in tumor immunology and viral entry studies. This recombinant human PVR construct, spanning residues 21–343 with an I340M substitution and a C-terminal hFc1 tag, has been validated by FACS to confirm direct binding to human TIGIT overexpressed on HEK293F cells, supporting use in receptor-ligand binding assays, TIGIT blockade screening, and functional cell-based checkpoint studies. Mammalian cell expression preserves native glycosylation patterns essential for proper folding of PVR's immunoglobulin-like domains, and the hFc1 tag facilitates detection in flow cytometry panels as well as surface plasmon resonance experiments. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based functional assays and in vivo immune checkpoint modeling.
