TNFSF9 (4-1BB ligand) plays a central role in T-cell co-stimulation and anti-tumor immunity, making it a critical reagent for immuno-oncology research. This tag-free recombinant human TNFSF9 (aa 71–254), expressed in *E. coli*, demonstrates potent bioactivity with an ED50 of less than 10 ng/mL in IL-8 stimulation of human PBMCs, corresponding to a specific activity exceeding 1.0 × 10⁵ IU/mg—providing reliable performance in PBMC activation assays, NF-κB signaling pathway studies, and T-cell co-stimulation experiments. Endotoxin levels below 1.0 EU/μg minimize LPS-driven artifacts that would otherwise confound cytokine readouts in sensitive immune cell functional assays. Purity exceeding 95% by SDS-PAGE, combined with this stringent endotoxin control, supports use as a reference standard in cytokine detection assays, a positive control for antibody validation, and a functional ligand in tumor biology animal models.
