SIGIRR, formally known as Single Ig IL-1-related receptor, is also referred to as Interleukin-1 receptor 8 (IL-1R8). Discovered and named by Thomassen et al. in 1999, SIGIRR emerges as a novel member of the Toll-IL-1R (TIR) signaling receptor superfamily. SIGIRR possesses a unique structural configuration, encompassing a single Ig extracellular domain, a transmembrane domain, an intracellular TIR conserved domain, and an intracellular tail region. In contrast to IL-1R and TLRs, SIGIRR features only one Ig extracellular domain, while IL-1R boasts three. Furthermore, the intracellular TIR domain of SIGIRR lacks the essential amino acids Ser447 and Tyr536 for signal transduction. SIGIRR exhibits the ability to suppress the signaling involved in T cell differentiation associated with IL-1 family members and TLR-mediated activation. This inhibitory capacity towards the signaling pathways of ILRs and TLRs family members renders SIGIRR a crucial regulatory factor in inflammation, tumor-associated inflammation, and autoimmunity. Extensive research has confirmed SIGIRR's negative regulatory role in ILRs and TLRs inflammatory signaling pathways, modulating relevant inflammatory responses in infectious diseases, tumors, and autoimmune disorders. Therefore, SIGIRR, as a novel anti-inflammatory molecule, holds potential in predicting the development of inflammation and immune-related diseases.
