TNFRSF18, also known as glucocorticoid-induced TNFR-related protein (GITR), is a crucial receptor molecule primarily expressed on the surfaces of cells involved in adaptive and innate immunity, including CD4+ T cells and CD8+ T cells. Structurally, TNFRSF18 exhibits similarities to other members of the tumor necrosis factor receptor superfamily, serving as a receptor capable of interacting with its ligands to trigger a series of biological effects. Functionally, TNFRSF18 primarily regulates the activity and function of T cells. Through its interaction with ligands, it influences various aspects of T cell activation, proliferation, and immune responses, while maintaining the balance of the immune system. In terms of signaling pathways, upon binding to its ligands, TNFRSF18 activates a cascade of signal transduction processes involving nuclear factor kappa B (NF-κB) and specific mitogen-activated protein kinase (MAPK) pathways, thereby modulating the activity and survival of T cells and affecting the strength and duration of immune responses. Additionally, the interaction between GITR and GITRL plays a pivotal role in immune regulation, enhancing immune responses and potentially serving as a therapeutic target for antitumor treatment. However, despite our current understanding of TNFRSF18, the detailed molecular mechanisms of its interaction with ligands and the involved signaling pathways still require further investigation. Future studies on TNFRSF18 are expected to provide a more comprehensive understanding of the immune system and theoretical support for its application in immunotherapy.
