Target Background

Function
Acts as catalytic component of the CCR4-NOT core complex, which in the nucleus seems to be a general transcription factor, and in the cytoplasm the major mRNA deadenylase involved in mRNA turnover. CCR4 has 3'-5' RNase activity with a strong preference for polyadenylated substrates and also low exonuclease activity towards single-stranded DNA. Discovered because of its role in the control of ADH2 gene expression. It is required for the expression of genes involved in non-fermentative growth and it mediates or is required for the action of the SPT6 and SPT10 genes.
Gene References into Functions
  1. Genome-wide mapping of decay factor-mRNA interactions identifies nutrient-responsive transcripts as targets of the deadenylase Ccr4. PMID: 29158339
  2. These results suggest that Ccr4 regulates not only LRG1 mRNA level through poly(A) shortening but also the translation of LRG1 mRNA, and that Pbp1 is involved in the Ccr4-mediated regulation of mRNA stability and translation. PMID: 28231297
  3. Collectively, these data demonstrate a novel role for Ccr4-Not in Pol I transcriptional regulation that is required for bridging mTORC1 signaling to ribosomal RNA synthesis. PMID: 25815716
  4. This review will describe what is known about the Ccr4-Not complex in regulating transcription elongation and its possible collaboration with other factors traveling with RNAPII across genes. PMID: 22975735
  5. Khd1 and Ccr4 modulate a signal from Rho1 in the cell wall by regulating the expression of RhoGEF and RhoGAP. PMID: 21873511
  6. report that four subunits of the Ccr4-Not complex-Ccr4p, Pop2p, Not2p, and Not4p-severely impact HSP104 RNA transcription site-associated retention induced by MFT1 gene deletion PMID: 21862638
  7. Ccr4-Pop2-NOT mRNA deadenylase contributes to septin organization; deletion of deadenylase subunits CCR4 or POP2, but not PAN2, resulted in septin morphology defects PMID: 19487562
  8. Combining spt15 point mutations (in TATA-binding protein TBP) with either mot1 or ccr4 results in either a growth defect or lethality. PMID: 16272410
  9. the Ccr4-Not4 complex controls ubiquitination of the nascent-associated polypeptide (NAC-EGD) complex PMID: 16926149
  10. Ccr4p catalyzes PUF protein-mediated deadenylation using a Pop2p bridge to recruit a multifunctional Pop2p complex to the mRNA PMID: 17090538
  11. CCR4/NOT complex associates with the proteasome and regulates histone methylation. PMID: 17389396
  12. These results support the model that the control of CCR4 deadenylation in vivo occurs in part through the removal of PAB1 from the poly(A) tail following its self-association into multimers and/or a circular species. PMID: 17620415
  13. the Ccr4-Not complex has a critical role in the mechanism of activation of Skn7 that is dependent upon the Srb10 kinase PMID: 17965252
  14. distinct portions of the CCR4-NOT complex control a number of different cellular processes PMID: 18214544
  15. Using RNA reporters that recapitulate Vts1p-mediated decay in vivo, study demonstrates that Vts1p stimulates mRNA degradation through deadenylation mediated by the Ccr4p-Pop2p-Not deadenylase complex. PMID: 18469165
  16. Ccr4 alters cell size in yeast by modulating the timing of CLN1 and CLN2 expression. PMID: 18493058
  17. Study revealed an impact of the Ccr4-Not complex on more than 85% of all studied genes, inferred that the Ccr4-Not complex is important for expression of most of the yeast genome and that each subunit had a particular impact on complex integrity. PMID: 19155328
