TNFRSF9 (4-1BB) is a costimulatory immune checkpoint receptor whose extracellular domain is a key target for agonist antibody development in immuno-oncology. This recombinant human 4-1BB construct spans residues 24–186 of the extracellular domain, expressed in mammalian cells with a C-terminal His-Fc tag to preserve native glycosylation and disulfide-dependent folding critical for accurate ligand and antibody recognition. Functional ELISA confirms binding activity with an ED50 of 0.41 µg/mL against an anti-human CD137 mAb, supporting use in therapeutic antibody epitope mapping, competitive blocking antibody screening, and affinity characterization by SPR or BLI. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/µg satisfies the criteria typical for cell-based co-stimulation assays and drug discovery screens targeting 4-1BB-mediated T-cell activation pathways.
