Presenilin enhancer gamma-secretase subunit (PSENEN), also known as PEN-2, is an essential regulatory component of the gamma-secretase complex, a multiprotein protease critical for intramembrane cleavage of substrates including amyloid precursor protein (APP) and Notch receptors. As a stabilizing subunit, PSENEN interacts with presenilin (PSEN1/PSEN2), nicastrin, and APH-1 to maintain γ-secretase activity, directly influencing the production of amyloid-β (Aβ) peptides implicated in Alzheimer's disease (AD) pathogenesis. Its catalytic function impacts Notch signaling, a pathway governing cell differentiation and oncogenesis. Pathogenic mutations in PSENEN are associated with familial acne inversa (AI) and rare early-onset AD cases. Drug development targeting γ-secretase has faced challenges due to its pleiotropic roles; however, selective γ-secretase modulators (GSMs) and Notch-sparing inhibitors remain under investigation for AD and cancer. Recent studies explore PSENEN's regulatory role in Aβ42/Aβ40 ratio modulation, a key therapeutic strategy against amyloidosis.

IHC image of CSB-PA878932OA01HU diluted at 1:400 and staining in paraffin-embedded human adrenal gland tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
