This ELISA kit provides quantitative measurement of human TP53 (p53) protein in research samples, supporting studies in cancer biology where p53 status directly influences tumor suppression, apoptotic signaling, and therapeutic response. Because p53 is predominantly an intracellular protein that accumulates upon DNA damage or stabilizing mutations, detecting it across both secreted and lysate-based matrices requires a kit designed for diverse sample inputs.
Detection Range: The 62.5–4000 pg/mL range captures the elevated p53 levels characteristic of cells harboring missense mutations that stabilize the protein, while accommodating the lower concentrations found in wild-type expressing tissues. Samples from tumors with high mutant p53 accumulation may require dilution to fall within range.
Sensitivity: At 31.25 pg/mL, the kit detects p53 in conditions where protein levels are low, such as wild-type p53-expressing cells under basal conditions or dilute cell culture supernatants containing released p53 following apoptotic events.
Sample Compatibility: Validation across serum, plasma, cell culture supernatants, cell lysates, and tissue homogenates enables researchers to correlate circulating p53 levels with intracellular accumulation in matched tumor and blood samples from the same subjects.
Assay Efficiency: The 1–5 hour protocol using only 50–100 µL accommodates precious clinical biopsies and primary tumor-derived cultures where material is limited.
