TNF-α drives pleiotropic inflammatory and apoptotic responses in both human and non-human primate models, making ortholog fidelity essential when translating findings from rhesus macaque studies to clinical contexts. This protein, spanning residues 77–233 of the mature macaque TNF sequence and expressed in mammalian cells, binds human TNFRSF1A with an EC50 of 9.1–10.0 ng/mL in functional ELISA, confirming receptor engagement at physiologically relevant concentrations and supporting its use in cross-species receptor-binding assays, NF-κB and MAPK signaling pathway studies, and as a positive control in cytokine detection platforms. Endotoxin levels below 1.0 EU/μg meet the thresholds commonly required for primary immune cell activation assays—including PBMC stimulation and T-cell differentiation experiments—where LPS contamination would confound TNF-specific responses. Purity exceeding 90% by SDS-PAGE, combined with the validated receptor interaction, provides a suitable basis for antibody development workflows and in vivo macaque inflammation models.
