CD69, officially named cluster of differentiation 69, is a type II C-type lectin protein also known as activation inducer molecule (AIM). As an early activation marker, it is rapidly expressed on the surface of lymphocytes, monocytes, and eosinophils upon activation, mediating cell adhesion and immune regulation through interactions with sphingosine-1-phosphate receptor 1 (S1PR1) to retain cells in lymphoid tissues.
In signaling pathways, CD69 modulates calcium flux and MAPK/ERK activation, influencing T-cell differentiation and cytokine secretion. Its dysregulation correlates with autoimmune diseases like rheumatoid arthritis and psoriasis, as well as lymphoproliferative disorders. Current research explores CD69-targeted antibodies for immunomodulatory therapy, with preclinical studies investigating their efficacy in suppressing overactive immune responses.
