Myelin-associated glycoprotein (MAG), also known as Siglec-4a, is a transmembrane protein predominantly expressed in the periaxonal Schwann cell and oligodendrocyte membranes, where it mediates glial-axonal interactions during myelination. Functioning as a sialic acid-binding immunoglobulin-like lectin (Siglec), MAG binds to gangliosides GD1a and GT1b on neuronal membranes, stabilizing myelin integrity and inhibiting axonal regeneration through RhoA/ROCK pathway activation. It interacts with the Nogo receptor (NgR) complex, synergizing with Nogo and OMgp to suppress neurite outgrowth in CNS injury. Autoantibodies against MAG are biomarkers for IgM paraprotein-associated demyelinating neuropathy, a subtype of chronic inflammatory demyelinating polyneuropathy (CIDP). Therapeutic strategies targeting MAG-NgR signaling, including anti-LINGO-1 antibodies and Rho kinase inhibitors, have shown limited efficacy in clinical trials, prompting exploration of combinatorial approaches for nerve repair.

Immunofluorescence staining of MCF-7 cells with CSB-PA013322LA01HU at 1:200, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
