DKK1

DKK1 encodes Dickkopf-1, a secreted glycoprotein and key Wnt signaling antagonist. It binds LRP5/6 co-receptors, blocking Wnt ligand interaction to inhibit β-catenin-mediated transcription. Dysregulated DKK1 is linked to osteoporosis via osteoblast suppression and bone metastases in cancers like multiple myeloma. Preclinical studies target DKK1 with monoclonal antibodies (e.g., BHQ880) to restore bone formation and suppress tumor growth, with early-phase trials evaluating efficacy in myeloma and bone disorders.
