STAT3 is a major member of the signal transduction and activator of transcription (STAT) family, and is a confluence of signaling pathways activated by many cytokines, growth factors, and some proto-oncogenes. Among of them, the JAK-STAT pathway is the most classical signal transduction pathway involving STAT3. About 50 cytokines can activate the JAK-STAT signaling pathway. In addition to JAKs, STAT3 is also phosphorylated by Src family kinases (Src). When some growth factors bind to cell surface receptors, resulting in the activation of Src, phosphorylation of STAT3, and activation of STATs. In addition to ligand binding-induced recruitment and activation of receptor-associated tyrosine kinases, there are also endogenous activation of non-receptor tyrosine kinases such as Abl and Src in the cytoplasm.
Under normal circumstances, STAT3 can act as the main regulator of the balance of cell proliferation and apoptosis to maintain the totipotent type of embryonic stem cells and participate in self-antigen tolerance and antigen presentation. Generally, its activation is controllable and temporary, and after completion of specific signaling, it is dephosphorylated and returned to the cytoplasm. However, under the stimulation of oncogenic signals, STAT3 will be continuously activated, and it will exist in the nucleus in an activated state, continuously activate target genes, and promote the growth of tumor cells.
Studies have confirmed that it acts as a regulator in the cycle of cell growth and immune response, and is highly expressed in most tumors. Therefore, many drug developers have been developing targeted drugs for STAT3. At present, many drugs have entered the clinic and achieved good responses, and there are more STAT3 inhibitors in preclinical development.
