Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

The expression region of this recombinant Escherichia coli (strain K12) spoT covers amino acids 1-702. The calculated molecular weight for this spoT protein is 86.8 kDa. The spoT protein was expressed in e.coli. The spoT coding gene included the N-terminal 10xHis tag and C-terminal Myc tag, which simplifies the detection and purification processes of the recombinant spoT protein in following stages of expression and purification.

Escherichia coli SpoT is a bifunctional enzyme involved in the stringent response, a regulatory mechanism that enables bacteria to adapt to environmental stress conditions. SpoT possesses (p)ppGpp synthase and hydrolase activities, allowing it to control the levels of guanosine pentaphosphate and tetraphosphate [(p)ppGpp], collectively known as alarmones. During nutrient limitation or stress, SpoT synthesizes (p)ppGpp, signaling a global shift in cellular processes, including a slowdown in growth and changes in gene expression. Conversely, SpoT can hydrolyze (p)ppGpp when conditions become more favorable. This dual functionality allows SpoT to finely tune the bacterial response to varying environmental cues, contributing to bacterial survival and adaptation strategies.

In eubacteria ppGpp (guanosine 3'-diphosphate 5'-diphosphate) is a mediator of the stringent response which coordinates a variety of cellular activities in response to changes in nutritional abundance. This enzyme catalyzes both the synthesis and degradation of ppGpp. The second messengers ppGpp and c-di-GMP together control biofilm formation in response to translational stress; ppGpp represses biofilm formation while c-di-GMP induces it. ppGpp activates transcription of CsrA-antagonistic small RNAs CsrB and CsrC, which downregulate CsrA's action on translation during the stringent response.
