CTLA4 is a gene providing an instruction of making a protein named cytotoxic T-lymphocyte protein 4 (also abbreviated as CTLA4), which is a transmembrane receptor on T cells. The protein named CTLA4, also known as CD152 (a leukocyte differentiation antigen), is an inhibitory receptor inducibly expressed following T cell activation and acts as a major negative regulator of T-cell responses. CTLA4 either binds to CD80 or CD86, leading to a costimulatory or a co-inhibitory response, respectively. Due to the dampening effect of CTLA4, it plays a crucial role in the regulation of T-cell homeostasis and self-tolerance.
Actually, the mechanism of CTLA-4 inhibiting T cells has not been elucidated completely. Based on some action mechanisms of CTLA-4 drugs, there are two main mechanisms of CTLA-4 inhibiting T cells.
One is CTLA-4 and APC. As mentioned before, CTLA-4 is a transmembrane receptor on T cells that shares a ligand named B7 with CD28. Activation of T cells requires co-stimulation by TCR/MHC and CD28/B7. CTLA-4 expressed on T cells can inhibit T cell function through binding of B7 molecules (including B7-1 and B7-2) on antigen presenting cells (APCs). Another is CTLA-4 and Tregs. As you know, CTLA-4 is abnormally highly expressed on the surface of tumor-infiltrating Treg cells, and the level of its expression is significantly higher than that of peripheral Treg cells and effector T cells in the periphery and tumor. Treg cells can inhibit over-activated T cells and other immune cells and plays a critical role in protecting the body. In terms of CTLA-4, treg cells can use granzyme A and granzyme B to express membrane molecules such as CTLA-4 in CTL and NK cells in a perforin-dependent manner, trans-endocytosing CD80 and CD86 molecules on DC cells, and weakening the antigen-presenting ability of DC cells.
