ERBB2

Receptor tyrosine-protein kinase erbB-2 (also known as ERBB2) is a protein encoded by ERBB2 gene. The protein encoded by this gene is also known as epidermal growth factor receptor 2 (HER2) and is a member of the epidermal growth factor receptor family (EGFR). ERBB2 protein is a transmembrane glycoprotein with tyrosine kinase activity, which has a molecular weight of approximately 185 kDa, also known as p185. HER2 plays a crucial role in tissue development and is essential for normal embryonic development. Notably, HER2 is usually under- or non-expressed in normal adult body tissues. Over-expressed HER2 often causes biological dysfunction, and many studies have shown that HER2 expression is closely related to the occurrence of many tumors.

Currently, no ligand has been identified for ERBB2. HER2 can heterodimerise with any of the other three receptors (such as HER3 or HER4) and is also considered to be the preferred dimerisation partner of the other ERBB receptors. HER2 dimerization results in a conformational change that activates intracellular tyrosine kinase activity, thereby initiating a variety of signaling pathways mainly including RAS-RAF-MEK-ERK for cell proliferation and PI3K-AKT-mTOR pathway for cell survival.

IHC image of CSB-RA007763MA2HU diluted at 1:600 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-Mouse IgG labeled by HRP and visualized using 0.05% DAB.
