IDH3, isocitrate dehydrogenase 3, is one of the key rate-limiting enzymes in the tricarboxylic acid cycle. It functions in the mitochondria of human cells, catalysing the conversion of isocitric acid to α-ketoglutarate with the simultaneous production of NADH and CO2.IDH3 consists of α, β, and γ subunits, which are assembled to form an α2βγ heterotetramer, and further to form an octamer. This structure provides IDH3 with efficient catalytic capacity and metabolic regulatory properties.The activity of IDH3 is variably regulated by a variety of metabolic small molecules, such as CIT, ADP, and ATP, which are able to modulate the activity of IDH3, and consequently affect cellular energy metabolism. In a biological sense, IDH3 is not only involved in cellular energy production, but may also be associated with cellular oxidative damage and lipid synthesis, and its mutation is directly related to the development of a variety of tumours, thus it has an important potential value in medical research and therapy.
