TNFRSF1B

TNFRSF1B, also called TNF-R2, is a receptor for TNF-α and is restricted to immune cells, endothelial cells, neural cells, and mesenchymal stem cells (MSCs). TNFR2 is mainly expressed on activated T cells and is involved in the regulation of immune responses through regulatory T cells (Tregs).

Besides THFR2, TNF also binds to a type I transmembrane receptors, TNFR1. Both TNFR1 and TNFR2 contain four cysteine-rich domains (CRDs) in their extracellular domains. The membrane-distal CRD contains the proligand-binding assembly domain (PLAD), which is important for ligand-mediated formation of active receptor complexes. In the absence of ligand, PLAD mediates receptor homomultimerization in an inactive self-crosslinked state.

TNF mainly recognizes TNFR1 for apoptosis and relies on TNFR2 for functions related to T cell survival. TNF-TNFR2 promotes the transcription of pro-survival genes through NF-κB signaling, thereby promoting cell proliferation and survival. Many experiments have shown that blocking antibodies can specifically kill Treg cells and tumor cells in the tumor microenvironment.
