TNF-α drives NF-κB and MAPK signaling cascades central to inflammation, apoptosis, and immune cell activation, making validated binding activity essential for meaningful downstream results. This recombinant human TNF (aa 77–233) demonstrates potent receptor engagement, with an EC50 of 3.470–4.107 ng/mL for human TNFR2 binding in functional ELISA, supporting its use as a coating antigen in antibody development, a positive control in cytokine detection assays, and a reference ligand in receptor-binding studies. Expressed in yeast with an N-terminal 6xHis tag, the protein covers the mature soluble domain and achieves greater than 90% purity by SDS-PAGE, which aligns with standards expected in cell-based activation and apoptosis assays; however, researchers should note that endotoxin levels have not been tested, so independent screening is advisable before use in sensitive immune cell experiments such as PBMC stimulation or primary T-cell proliferation studies.
Applications : immunofluorescence staining
Review: Control NP and AF cells compared with those exposed for 12 hrs to TNF (25 ng/ml), TNF+CM, TNF+EVs or TNF+ SF. Localization of NF-κB p65 was visualized using confocal microscopy after immunofluorescence staining with NF-κB p65 antibody (green). Nuclei (white arrows) were stained with DAPI (blue).

Applications : Thermos shift assay
Review: Thermal shift curves of unfolding transition of TNF-alpha in the presence of 100 pM, 10 pM, 1 pM, 0.1 pM 0.01 pM, and 0 pM of MY1. Data fit Boltzmann equation gave midpoint Tm of 81.13°C, 78.52°C, 77.07.5°C, respectively, (R2 > 0.9).
