TRPV6, the transient receptor potential channel family, Vanilloid subfamily member 6", alias ECACAC1. it is a membrane protein belonging to the TRP cation channel family and is expressed predominantly in epithelial tissues of the small intestine, kidneys, placenta, epididymis, and certain secretory glands. the mechanism of action of TRPV6 involves the regulation of intracellular calcium ions ( Ca2+) regulation and is involved in the selective permeation of calcium ions through its channel activity, which is essential for the maintenance of calcium homeostasis and physiological functions in vivo.The TRPV6 gene is a direct transcriptional target of the active form of vitamin D, and its expression is regulated by the level of vitamin D in response to the body's requirement for calcium ions. In addition, TRPV6 may also fulfill its function by interacting with other proteins in the membrane such as calmodulin.
The biological significance of TRPV6 is mainly in its regulation of calcium ion homeostasis. It is particularly important for its function in the intestine and kidney, where it is involved in the process of calcium ion absorption and reabsorption, thus helping to maintain blood calcium levels.Loss-of-function mutations in TRPV6 have been associated with a number of disorders, such as hypercalciuria, decreased bone mineral density, insufficient body weight gain, reduced fertility, and even alopecia. In addition, TRPV6 has been found to be overexpressed in some malignant tumors, and its upregulation is closely associated with tumor aggressiveness, metastasis, and poor survival, suggesting that TRPV6 may play a contributory role in tumor development. Thus, TRPV6 not only plays a key role in maintaining physiological functions, but may also be a potential target for the treatment of certain diseases, especially cancer therapy.

IHC image of CSB-PA861992LA01HU diluted at 1:300 and staining in paraffin-embedded human placenta tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
