Target Background

Function
(From Uniprot)
Plasma membrane-associated small GTPase which cycles between active GTP-bound and inactive GDP-bound states. In its active state, binds to a variety of effector proteins to regulate cellular responses such as secretory processes, phagocytosis of apoptotic cells, epithelial cell polarization, neurons adhesion, migration and differentiation, and growth-factor induced formation of membrane ruffles. Rac1 p21/rho GDI heterodimer is the active component of the cytosolic factor sigma 1, which is involved in stimulation of the NADPH oxidase activity in macrophages. Essential for the SPATA13-mediated regulation of cell migration and adhesion assembly and disassembly. Stimulates PKN2 kinase activity. In concert with RAB7A, plays a role in regulating the formation of RBs (ruffled borders) in osteoclasts. In podocytes, promotes nuclear shuttling of NR3C2; this modulation is required for a proper kidney functioning. Required for atypical chemokine receptor ACKR2-induced LIMK1-PAK1-dependent phosphorylation of cofilin (CFL1) and for up-regulation of ACKR2 from endosomal compartment to cell membrane, increasing its efficiency in chemokine uptake and degradation. In neurons, is involved in dendritic spine formation and synaptic plasticity. In hippocampal neurons, involved in spine morphogenesis and synapse formation, through local activation at synapses by guanine nucleotide exchange factors (GEFs), such as ARHGEF6/ARHGEF7/PIX. In synapses, seems to mediate the regulation of F-actin cluster formation performed by SHANK3. In neurons, plays a crucial role in regulating GABA(A) receptor synaptic stability and hence GABAergic inhibitory synaptic transmission through its role in PAK1 activation and eventually F-actin stabilization.
Gene References into Functions
  1. Live cell imaging with cell cycle indicators showed that expression of Rac1V12, but not the wild-type Rac1 or Rac1b, promoted cell cycle progression. The expression of Rac1b per se cannot induce cell proliferation. PMID: 23411476
  2. Silencing tight junction protein ZO-2 alters epithelial cytoarchitecture.ZO-2 silencing increases RhoA activity through GEF-H1 recruitment to cell borders.ZO-2 silencing activates Cdc42 and triggers mitotic spindle disorientation. ZO-2 silencing increases Rac1 and cofilin activity and lamellipodia formation. PMID: 28554775
  3. Data indicate exosomal Rac1/PAK2 as angiogenic promoters. PMID: 26919098
  4. This inversion of glandular orientation is accompanied by Rac1 inactivation during early cystogenesis, and temporal activation of Rac1 is sufficient to recover the normal cyst phenotype. PMID: 23051733
  5. These results indicate that cadherins and nectins cooperate to induce and then rapidly suppress Rac1 activity during initial cell-cell adhesion. PMID: 21412440
  6. Numb acts to suppress Rac1-GTP accumulation, and its loss leads to increased sensitivity toward extracellular signals that disrupt cell-cell adhesion to induce epithelial-mesenchymal transition (EMT) and cell dispersal PMID: 21147098
  7. Rac activation by cadherin through the c-Src-Rap1-phosphatidylinositol 3-kinase-Vav2 pathway PMID: 16170364
  8. these results provide the first evidence for a role of Rac1/Cdc42 signalling during early HSV-1 infection and suggest a mechanism relying on virus-induced regulation of Rac1/Cdc42 activities. PMID: 17098962
  9. ARF6 regulates the subcellular distribution of the GTPase, Rac1, to tubule extensions. ARF6-induced ERK activation regulates Rac1 activation during tubule initiation through the expression of the receptor for urokinase type plasminogen activator. PMID: 17363898
  10. Data show that zones of Rac1 and lamellipodia activity and of RhoA and actomyosin contractility are restricted to the periphery of contacting membranes and together drive initiation, expansion, and completion of cell-cell adhesion. PMID: 17646397
  11. Phosphoinositide 3'-kinase is required for apical membrane protein remodeling from the initial apical membrane surface. Both Rac1 and atypical PKC are required for reorientation of cellular polarity. PMID: 17804488
  12. phosphatidylinositol 3-kinase and Rac1 are transiently activated by E-cadherin engagement and initiate a new round of membrane dynamics, but they are subsequently suppressed at that site to allow maintenance of weak E-cadherin mediated adhesion. PMID: 18089563
  13. Paracingulin regulates the activity of Rac1 and RhoA GTPases by recruiting Tiam1 and GEF-H1 to epithelial junctions PMID: 18653465
  14. as a mediator of SF- and Src-stimulated NF-kappaB activity. Finally, the Src/Rac1/MKK3/6/p38 and Src/TAK1/NF-kappaB-inducing kinase pathways exhibited cross-talk at the level of MKK3. PMID: 19047046
