ALK, or Anaplastic Lymphoma Kinase", alias Anaplastic Lymphoma Kinase, CD246. it is a receptor tyrosine kinase, encoded by its own genes in human cells, which is mutated in a wide range of malignant tumors.ALK fusion occurs at a frequency of 3-7% in NSCLC, and most of them are the EML4-ALK fusion form; ALK secondary resistance mutations account for approximately 37% of drug resistance in ALK-positive NSCLC, respectively, mutations in the ALK kinase region (28%) and copy number amplification of the ALK fusion gene (9%).The mechanism of action of ALK involves its function as a cell-surface receptor, which, when bound to a specific ligand, dimerizes and autophosphorylates ALK, activating downstream signaling pathways. These signaling pathways include PI3K/AKT, MAPK, JAK/STAT, etc., which play important roles in cell proliferation, differentiation, migration and apoptosis. In tumor cells, aberrant activation of ALK can promote tumorigenesis and development.
ALK has a wide range of biological significance, which not only participates in the normal physiological process of cells, but also is closely related to the development of a variety of diseases. In tumor cells, abnormal activation of ALK can promote tumorigenesis and development, especially in non-small cell lung cancer, where the presence of ALK fusion genes is an important therapeutic target. Inhibitors targeting ALK, such as crizotinib, have been developed and used in the clinical treatment of ALK-positive non-small cell lung cancer patients, significantly improving the survival and quality of life of patients.
