BACH1

The BACH1 protein, encoded by the BACH1 gene, is a basic region-leucine zipper (bZIP) transcription factor belonging to the Cap'n'Collar (CNC) family. It is also known by aliases such as BACH1a and hBACH1. BACH1 functions as a transcriptional repressor by heterodimerizing with small Maf proteins and binding to antioxidant response elements (AREs) in target gene promoters, thereby inhibiting the expression of genes involved in oxidative stress responses and heme metabolism.
BACH1 is involved in regulating signaling pathways such as the Nrf2-ARE pathway, where it competes with Nrf2 for ARE binding to modulate cytoprotective gene expression. Dysregulation of BACH1 has been linked to various diseases, including cancer (e.g., breast, lung, and pancreatic cancer) through promotion of angiogenesis and metastasis, as well as cardiovascular diseases. Recent drug研发 efforts focus on developing small-molecule inhibitors targeting BACH1 to enhance antioxidant defenses and suppress tumor progression, with some compounds showing promising preclinical results.
