BTK, Bruton's tyrosine kinase, alias AGMX1 ATK BPK, is a non-receptor tyrosine kinase, a member of the Tec kinase family, which is widespread in all hematopoietic cells, except T cells and plasma cells.The mechanism of action of BTK is mainly to deliver the signals received by BCR and to regulate B-cell proliferation and survival through the participation in the B-cell receptor (BCR) signaling pathway. BTK contains five structural domains: PH, TH, SH3, SH2 and SH1/TK catalytic domains, which can recognize and bind to a variety of signaling molecules.
BTK has a wide range of biological significance, which not only participates in the maturation, survival, differentiation, apoptosis, migration and other processes of B cells, but also involves in the development of a variety of diseases, especially in the development of B cell lymphoma. In B-cell lymphoma, BTK is a key kinase in the BCR signaling pathway.BTK inhibitors can covalently bind to the cysteine residues in the active center of BTK, thereby inhibiting its activity and thus inhibiting the activation of the BCR signaling pathway, effectively preventing the migration of tumors from B-cells to lymphoid tissues that are suitable for tumor growth, reducing the malignant proliferation of B-cells, and inducing apoptosis of the cells, and thus exerting their therapeutic effects.

IHC image of CSB-RA553167A0HU diluted at 1:100 and staining in paraffin-embedded human tonsil tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
