CBLIF, also known as gastric intrinsic factor (IF) or GIF, is a glycoprotein secreted by gastric parietal cells. It specifically binds to dietary vitamin B12, protecting it from degradation in gastric acid and facilitating its absorption in the ileum, which is essential for erythropoiesis and neurological function.
Defects in CBLIF cause congenital intrinsic factor deficiency and pernicious anemia, leading to megaloblastic anemia and cognitive impairment. Current treatment relies on vitamin B12 supplementation. Recombinant CBLIF and absorption-enhancing formulations are under preclinical development to improve oral delivery for patients with malabsorption disorders.
