ENGASE, endo-beta-N-acetylglucosaminidase, alias endo-beta-N-acetylglucosaminidase. It is a human genetically encoded cytoplasmic enzyme whose primary function is to catalyze the hydrolysis of peptides and proteins with mannose modifications to produce free oligosaccharides.
ENGASE specifically hydrolyzes the β-1,4 glycosidic bond between two N-acetylglucosamines on the glycan chain in N-glycoproteins and belongs to the glycoside hydrolase 85 family; in addition to its hydrolytic activity, it also possesses a transglycosylation activity, which enables it to transfer the N-glycan chain to glycoproteins bearing a GlcNAc glycoside.ENGASE plays an important role in the process of protein deglycosylation, and the abnormal activity may be associated with certain congenital disorders of impaired deglycosylation. These functions of ENGASE are essential for the maintenance of protein structure and function, cell signaling, and normal physiological activities of organisms.
